Exercise 4

Due: microscope handout

YOU MUST USE COLORED PENCILS

This lab is to allow you to become familiar with the function & structure of microscopes.

Procedure

  1. Use a slide with a letter on it.  Which direction does the letter point?  Try moving it around a bit. Figure out why the image is upside down and backwards and change powers. Draw what you see in the upper 3 boxes.
  1. Use your slide with the letter and determine the affect of working distance, field of view, and parfocal capabilities on your ability to see your slide.
  1. Make 2 wet mounts of your cheek cells.  Stain one and leave the other alone.  Compare them and draw a picture of each slide in the bottom 2 boxes.
  1. Look at and draw 2 other tissue slides and the 3 different powers. Use color pencils.  Look for the structure and shape of each individual cell

For the first practical you will be responsible for:

  1. Identify the parts of a microscope.

base

stage clips

 fixed light source with dimmer.

arm

mechanical stage knobs

condenser

stage

course focus knob

iris diaphragm lever

fine focus knob

ocular lens

objective lens

nosepiece

scanning lens

high power lens

  1. Calculate the total magnification.

Total magnification =the power of the objective lens  x  the power of the ocular lens

  1. Be able to trouble shoot major problems

working distance

field of view

resolution

parfocal capability

  1. Describe why tissues are stained (Not the types so much as the reasons for the stains)
